Protein intact mass analysis involves directly detecting the precise molecular weight of antibody/recombinant proteins using mass spectrometry in a state where the protein is intact and not enzymatically cleaved into peptides. This is used for sequence confirmation, post-translational modification characterization, isoform differentiation, and batch consistency quality control. It is a core analysis for primary structure characterization in biologics, and for ADC drugs, it can determine the DAR value. Antibody intact mass analysis services mainly include non-glycosylated intact mass analysis, glycosylated intact mass analysis, glycosylated reduced light and heavy chain subunit intact mass analysis, and Fab and Fc mass analysis.

Analysis Objective

(1) Confirm whether the theoretical intact mass of the protein is consistent with the measured intact mass and verify the correctness of the expression sequence;

(2) Identify the heterogeneity of modifications such as glycosylation, glycation, oxidation, and C-terminal lysine deletion;

(3) Distinguish between the main peak, modified variants, truncated variants, and incomplete processing impurities.
