Overview: BLI (Bio-layer Interferometry) utilizes fiber optic biosensors to detect changes in the thickness of the sensor's optical layer during molecular binding and dissociation in real time. When biomolecule A binds to the sensor tip, it forms a biofilm. When molecule A binds to molecule B, it causes a change in the molecular weight at the sensor tip, resulting in a change in the biofilm thickness. Light passes through the biofilm layer of the sensor, undergoing transmission and reflection to form interference waves. The change in biofilm thickness causes a relative shift in the interference waves. The interference waves before and after biomolecule binding are detected by a spectrometer, forming an interference spectrum, which is displayed as the real-time shift (nm) of the interference pattern. Finally, the molecule to be detected is analyzed based on the changes in the spectrum before and after molecular binding, making it widely applicable to biomolecular interaction analysis.
