Product Overview

SEC (Size Exclusion Chromatography), also known as gel filtration chromatography, uses porous gel particles as packing material. Larger molecules have a greater hydrodynamic radius, making it more difficult for them to enter the micropores of the packing material, resulting in shorter elution times; smaller molecules enter the micropores more easily, but the longer the elution path, the later the elution. Separation is entirely based on molecular size/hydrodynamic radius, without relying on polarity, charge, or affinity. Antibody drugs, primarily IgG monomers, aggregates, and fragments, exhibit significant molecular weight variations, making them highly suitable for SEC separation.

Analysis Objective

(1) Antibody monomer: active main component, effective component of drug

(2) High molecular weight aggregate (HMW): dimer, polymer, oligomer

(3) Low molecular weight degradation products (LMW): antibody fragments, cleavage fragments, haptens, degradation products
